Plasmonic nanoparticles for optical read-out in applications in the visible wavelength range.

Gold nanoparticles exhibit an intensive red colour in aqueous dispersions caused by the so called localized surface plasmon resonance (LSPR). Light of a specific wavelength range enhances the characteristic frequency of the particles surface electrons, which undergo a collective oscillation (plasmon resonance). This phenomenon extremely amplifies the absorption of visible light of gold nanoparticles. A few femtomol of these particles on surfaces is sufficient for a visible detection. Furthermore, the strong light scattering of gold nanoparticles can be exploited e.g. in darkfield- or confocal microscopy techniques.

Citrate stabilized gold nanoparticles exhibit a strong affinity to molecules with positive net charge or positively charged functional groups, respectively. In particular, thiol groups bind as strong as quasi-covalent to gold nanoparticles.

The extremely high extinction coefficient in combination with the ease of biofunctionalization renders CANdot Series G ideally for the generation of selective molecular markers (up to 5 orders of magnitude higher sensitivity than fluorescence-based detection techniques) e.g. for use in immunoassays. A quantitative assay read-out via absorption intensity is feasible, according to Lambert-Beer’s law. Alternatively, CANdot ® Series G are offered pegylated, either neutral or with amine- or carboxyl groups for facile surface modifications.
